Johnson & Johnson Clinical Trials Show Progress Across Key Therapies

Johnson & Johnson (NYSE:JNJ) announced positive findings from key clinical evaluations testing its therapies, Tremfya and Carvykti, across psoriatic arthritis and multiple myeloma.

Tremfya Achieves Key Endpoints In Psoriatic Arthritis Study

In the Phase 4 STAR trial, Tremfya met its primary and main secondary endpoints among biologic-naive adult patients with active psoriatic arthritis presenting axial involvement.

The study prospectively evaluated an IL-23 inhibitor using magnetic resonance imaging to confirm axial disease.

At Week 24, patients receiving Tremfya demonstrated notable improvement in composite disease activity scores measuring spinal pain, morning stiffness, fatigue, and joint symptoms compared to placebo.

The therapy also reduced axial symptoms and decreased sacroiliac joint inflammation. The overall safety profile matched earlier evaluations, showing no new safety signals.

Axial involvement affects the spine and sacroiliac joints, causing mobility loss and severe pain in 5% to 28% of early psoriatic arthritis patients and up to 70% of long-standing cases.

Tremfya remains the sole fully human, dual-acting monoclonal antibody targeting IL-23 and CD64, holding regulatory approval to halt structural joint damage progression.

Carvykti Delivers Five-Year Progression-Free Remissions

Separately, long-term follow-up from Cohort A of the Phase 2 CARTITUDE-2 study revealed that 50% of relapsed or refractory multiple myeloma patients treated with a single Carvykti infusion remained alive and progression-free at five years without ongoing maintenance therapy.

The cohort evaluated 20 patients with one to three prior treatment lines who were refractory to lenalidomide.

At a median follow-up of 60.7 months, overall survival reached 69.2%, while median progression-free survival stood at 60.5 months. Among three patients evaluated for minimal residual disease at five years, all were negative at the deepest level tested.

Long-term remission occurred even in individuals with high-risk cytogenetic features.

Safety findings aligned with prior data, showing no new CAR T-cell-related neurotoxicity. One patient developed acute myeloid leukemia, and two deaths resulted from progressive disease and new cancer.
