# Nature Aging: Clinical research shows that supplementing with ω-3 fatty acids can delay aging, and the effect is even better when combined with vitamin D and exercise

Previous clinical studies have shown that restricting caloric intake may slow human aging. Other animal studies or small pilot trials have also indicated that supplementation with vitamin D or ω-3 has good prospects for slowing biological aging. However, it remains unclear whether these interventions are effective for humans.

Clinical scientists Heike Bischoff-Ferrari from the University of Zurich and renowned anti-aging researcher Steve Horvath from Altos Labs take ω-3 and vitamin D supplements daily to prevent aging-related health issues.

Their practice stems from a study they recently published. On February 3, 2025, they published a paper titled: *Individual and additive effects of vitamin D, omega-3 and exercise on DNA methylation clocks of biological aging in older adults from the DO-HEALTH trial* in the journal *Nature Aging*.

This clinical study indicates that taking ω-3 and vitamin D supplements over three years can delay biological aging by three to four months, and the effect is even better when combined with exercise.

ω-3 (omega-3) polyunsaturated fatty acids are essential nutrients for the human body. Since the body cannot synthesize them on its own, they must be obtained through food. ω-3 mainly includes alpha-linolenic acid (ALA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA), commonly found in deep-sea fish (such as salmon, trout, herring) and some plants (such as algae, walnuts, soybeans). Of course, they can also be obtained more simply and directly through supplements.

In this study, the research team analyzed data from the DO-HEALTH trial, which studied the effects of supplements and exercise on older adults and was conducted in five European countries from 2012 to 2014. The research team reviewed data from 777 participants aged 70 and older, who were divided into the following eight groups: placebo group, vitamin D group, ω-3 group, simple home exercise (SHEP) group, vitamin D + ω-3 group, vitamin D + SHEP group, ω-3 + SHEP group, and vitamin D + ω-3 + SHEP group The dosage of vitamin D is 2000 international units (IU) per day, the dosage of ω-3 is 1 gram per day, and the amount of exercise is 30 minutes of simple home exercise three times a week. About half of the participants were healthy, without significant chronic diseases or disabilities.

The research team collected blood samples from participants before and after the study and analyzed them using four different epigenetic clocks (PhenoAge, GrimAge, GrimAge2, and DunedinPACE). These epigenetic clocks measure biological aging through the methylation levels in DNA, and using several different epigenetic clocks allows for a more comprehensive capture of different aspects of biological aging.

The results showed that taking ω-3 supplements could slow down the aging rate of three epigenetic clocks (PhenoAge, GrimAge2, and DunedinPACE), and this effect was independent of the participants' gender, age, or body mass index. Additionally, combining vitamin D, ω-3, and exercise had extra benefits for slowing the aging of the PhenoAge epigenetic clock, reducing participants' biological aging by 2.9-3.8 months over three years.

Furthermore, the study found that the combination of vitamin D, ω-3, and exercise had the greatest impact on reducing cancer risk and preventing frailty. The research team believes that each intervention works through different but related mechanisms, and when combined, they reinforce each other, resulting in a stronger overall effect.

The research team stated that slowing biological aging by three to four months may seem small, but it can translate into significant public health benefits, such as reducing the incidence of some age-related health issues.

Overall, this clinical study indicates that ω-3 supplements have a protective effect in slowing biological aging, and this protective effect is reflected across multiple epigenetic clocks. Additionally, assessments based on the PhenoAge epigenetic clock show that ω-3, vitamin D, and exercise have an additive protective effect in slowing biological aging.

It is worth mentioning that the team published a research paper titled: *Combined Vitamin D, Omega-3 Fatty Acids, and a Simple Home Exercise Program May Reduce Cancer Risk Among Active Adults Aged 70 and Older: A Randomized Clinical Trial* in the journal *Frontiers in Aging* in April 2022 The study shows that combining vitamin D, ω-3, and exercise can reduce the cancer risk in seniors over 70 by 61%
