In the completed Phase III clinical studies (SYMPHONY-1&2), HTD1801 significantly improved renal function in patients with type 2 diabetes (T2DM) whose baseline eGFR was 60–90 mL/min/1.73m². After 52 weeks of treatment, the eGFR of these patients increased by an average of +3.08 mL/min/1.73m² (95% CI: 0.46–5.70), with no occurrence of hyperfiltration or fluid retention. These results suggest that HTD1801 has differentiated advantages over existing mechanism drugs and possesses the potential to delay or even reverse disease progression.

This study further explains the aforementioned clinical results. Conducted jointly by Haisco Pharmaceutical and the research team of Academician Jiang Jiandong from the Institute of Medicinal Biotechnology, Chinese Academy of Medical Sciences, the results show that in glucose and palmitic acid-induced podocyte injury models, HTD1801 can significantly increase podocyte survival rate and inhibit apoptosis; restore the expression of key podocyte structural proteins nephrin and podocin; and significantly reduce the levels of phosphorylated NF-κB in the metabolic inflammation pathway and the apoptosis execution protein caspase-3. In a diabetic nephropathy (DN) model, HTD1801 improved renal tissue structure in a dose-dependent manner, reduced the renal tubular injury score, alleviated renal inflammation and fibrosis lesions, and significantly lowered 24-hour urinary microalbumin levels.

This study systematically elucidates that HTD1801 can inhibit podocyte inflammation and apoptosis, stabilize glomerular structure, further revealing the kidney protection potential of HTD1801, providing important scientific evidence for its potential as a therapy for chronic kidney disease (CKD) and other kidney diseases.
